Curcumin

Curcumin nanocrystals
Curcumin is a polyphenolic compound extracted from turmeric that has been widely used for centuries as a food coloring, spice, or traditional herbal remedy. It is an approved food additive by the World Health Organization, the U.S. Food and Drug Administration, and numerous other countries, and is also an approved feed additive in China.
Modern pharmacological studies have demonstrated that curcumin exhibits a broad spectrum of pharmacological activities, including anti-inflammatory, antioxidant, antitumor, antiviral, anti-infective, anticoagulant, lipid-modulating, and cardioprotective effects, with low toxicity and few adverse reactions. However, its poor water solubility (less than 1 ppm in water) and low bioavailability severely limit its therapeutic efficacy, which has thus far prevented its approval as a pharmaceutical drug. Instead, it is currently used only in food and dietary supplements, thereby restricting its clinical application.

Curcumin nanocrystals
Based on the principles of mechanochemistry and condensed-matter physics, our company has pioneered the preparation of curcumin nanocrystals using a unique hot-melt extrusion process. Through a fully innovative extrusion-based crushing and nano-dispersion mechanism, curcumin is uniformly dispersed in high-performance excipients in the form of nanoscale microcrystals. Upon oral administration, this formulation significantly increases the contact area with gastrointestinal fluids, enabling rapid dissolution and absorption, thereby markedly enhancing the bioavailability of curcumin, reducing the impact of dietary factors on drug absorption, and improving the safety and reliability of therapeutic outcomes.
This product is formulated with pharmaceutical excipients that are safe, non-toxic, and exhibit excellent biocompatibility, making it suitable for high-demand routes of administration such as injection and inhalation. Compared with currently available commercial products, this product offers the following advantages:
Fast dissolution rate
Curcumin is highly dispersed in the carrier material as nanoscale particles, resulting in faster dissolution and higher solubility compared with conventional formulations.

High bioavailability
Nanocrystals not only enhance the dissolution rate and solubility of curcumin but also increase its adhesion to the gastrointestinal mucosa, thereby prolonging its residence time in the gastrointestinal tract and resulting in higher bioavailability. Following oral gavage administration in rats, the nanocrystal formulation exhibited a Cmax (maximum plasma concentration) 34-fold greater and an AUC (area under the plasma–time curve) 29-fold greater than the conventional drug substance.

Good stability
Curcumin exists in crystalline form in a low-energy state, exhibiting no polymorphic transformation or aging; consequently, it demonstrates superior stability compared with amorphous curcumin formulations prepared via conventional solid dispersion techniques.

Good security
Curcumin nanocrystals prepared using a small amount of biocompatible, pharmaceutically acceptable excipients as carriers do not alter the physiological functions of the gastrointestinal mucosa and exhibit higher safety compared with products that rely on solubilizing or co-solubilizing agents. Acute toxicity studies demonstrated that when rats were administered 5 g of curcumin nanocrystals per kilogram of body weight by gavage for two weeks, they exhibited normal growth and no adverse reactions were observed.
A long-term toxicity study [1] demonstrated that, in two dose groups—high and low—animals were administered the test substance continuously for 80 days, with some animals remaining under observation for an additional two weeks after drug cessation. The assessment included general condition monitoring, measurement of 10 hematological biochemical parameters and 8 hematological indices, as well as gross anatomical and histopathological examinations. The results showed that, compared with the normal control group, there were no significant differences in any of the measured parameters in either the high-dose group (500 mg/(kg·d)) or the low-dose group (100 mg/(kg·d)), indicating that continuous administration of curcumin at the aforementioned doses for 80 days is safe. 1. Long duration of action
Compared with products prepared using other solubilization techniques, nanocrystal formulations can maintain a constant absorption rate over an extended period, resulting in more stable plasma drug concentrations and a longer duration of action.
